Tissue Culture Studies in Pigeonpea

Abstract


Among the 11 explants used, explants like whole seed (Soaked and Unsoaked) middle and distal portions of cotyledon, leaf bits from matured leaves, corolla did not respond to callus induction. MS medium supplemented with 2, 4-D (2.0 mg/l) alone or with Kinetin (0.5 or 1.0 mg/l) was found to be suitable for callus induction from dehusked seed, proximal end of cotyledon, root tip, epicotyl segments. The combination of 2, 4-D (0.5 mg/l) NAA (1.0 mg/l) and BAP (2.0 mg/l) was suitable for callus induction from leaf bits of first pair of leaves. Shoot regeneration was achieved from the calli derived from dehusked seed and proximal end of cotyledon on medium with NAA (0.5 mg/l) and BAP (2.0 or 3.0 mg/l). Group of condensed shoots were also produced from dehusked seed on medium containing NAA (1.0 mg/l) along with BAP (3.0 mg/l) without callus stage.
