Effect of external damage on regeneration of cotton explants (Gossypium arboreum and G. barbadense)

Abstract


Several methods of injuring the plant for transformation and regeneration were examined in an effort to improve the efficiency of production of transgenic cotton. Agrobacterium tumefaciens strain EHA-105 carrying cry 1F was used for transformation study. The effect of wounding on established seedling, effect of vertical cut and effect of horizontal cut on well established seedling and regeneration was studied. Growth of plants was severely affected when injury made to remove shoot apical meristem (SAM) treatments. The putative transgenic plant was obtained when SAM was subjected to vertical cut.
