Optimization of growth conditions for production of saccharolytic enzymes by cellulolytic fungi

Abstract


Five cellulolytic fungi were screened for saccharolytic enzyme production. The growth of cellulolytic fungi at 300C temperature and pH 6.0 with cellulose as the carbon source over a period of 8 days was found to be ideal for secretion ofmaximum cellulase enzyme in all the fungi studied. Whereas for the activity of crude cellulase enzymes, 500C reactiontemperature with citrate buffer of pH 4.8 (0.05 M), T. reesei showed highest FPU activity (2.23 U ml-1), CMCase activity(3.68 U ml-1), â-glucosidase activity (1.82 IU ml-1) on cellulose and xylanase activity (3.65 U ml-1) using sugarcane bagasse as carbon source. The other best fungi crude enzyme extract observed was from T. viridae with 2.21 U ml-1 of FPU activity, 2.90 U ml-1 CMCase activity, 1.56 IU ml-1 â-glucosidase activity and 2.75 U ml-1 xylanase activity on cellulose as carbon source when compared to Aspergillus sydowii, Aspergillus awamorii and Phanerochaete chrysosporium.
